Senna leaf (Senna alexandrina) is a widely used stimulant laxative approved in many pharmacopoeias, with sennosides A and B as its primary active compounds. It is clinically effective for constipation management and bowel preparation, but prolonged use can lead to dependency and melanosis coli.
Sennosides are converted by gut microbiota into rheinanthrone, which stimulates colonic peristalsis via activation of prostaglandin E2 and nitric oxide pathways, increasing fluid secretion and motility. The compounds also inhibit Na+/K+ ATPase, reducing water absorption, and may interact with serotonin receptors to enhance propulsive activity.
