Ayahuasca Admixture Chacruna (Psychotria viridis): Benefits, Dosage, Safety & Evidence

Frequently asked questions

How does chacruna work in ayahuasca brews?

Chacruna (Psychotria viridis) contains N,N-dimethyltryptamine (DMT), a serotonergic psychedelic compound. DMT is normally broken down by monoamine oxidase enzymes in the digestive tract before reaching the brain. When combined with Banisteriopsis caapi, which contains beta-carboline MAO inhibitors like harmine, DMT becomes orally active and crosses the blood-brain barrier. This synergistic mechanism—DMT providing the visionary effects while MAO inhibitors enable oral absorption—is fundamental to ayahuasca's psychoactive properties. (Evidence Level: C)

Can chacruna interact with antidepressants or MAO inhibitors?

Chacruna poses serious drug interaction risks, particularly with antidepressants. Combining DMT-containing plants with SSRIs, SNRIs, or pharmaceutical MAO inhibitors can trigger serotonin syndrome, a potentially life-threatening condition causing agitation, hyperthermia, rapid heart rate, and seizures. Additionally, psychiatric medications may unpredictably alter ayahuasca's psychological effects. Individuals taking antidepressants require medical supervision and an appropriate washout period before any exposure. (Evidence Level: C)

Is chacruna safe during pregnancy or breastfeeding?

Chacruna is considered unsafe during pregnancy and breastfeeding. DMT crosses the placental barrier and passes into breast milk, potentially affecting fetal and infant neurological development. Traditional indigenous practices typically exclude pregnant and nursing women from ayahuasca ceremonies. The intense psychological experience and physiological stress also pose risks to maternal health. Women who are pregnant, planning pregnancy, or breastfeeding should avoid chacruna entirely. (Evidence Level: C)

What is the traditional dosage of chacruna in ayahuasca preparation?

Traditional ayahuasca preparations typically use 25-50 grams of fresh chacruna leaves combined with Banisteriopsis caapi vine. However, potency varies significantly based on plant source, preparation method, and individual sensitivity. Dosage should only be determined by experienced practitioners within ceremonial contexts. Self-administration is strongly discouraged due to unpredictable psychological effects and potential adverse reactions. There is no established safe dosage for unsupervised use. (Evidence Level: Traditional)

What are the side effects and risks of chacruna in ayahuasca?

Chacruna-containing ayahuasca produces intense psychedelic experiences including vivid visual hallucinations, altered perception of time and space, and profound emotional processing. Physical effects commonly include nausea, vomiting (traditionally considered purgative and therapeutic), and elevated heart rate. Psychological risks include anxiety, panic reactions, and potential exacerbation of underlying psychiatric conditions. Individuals with cardiovascular disease, schizophrenia spectrum disorders, or taking serotonergic medications face elevated risk and should avoid use. (Evidence Level: C)

Summary

Psychotria viridis (Chacruna) is a shrub from the Amazon rainforest used as a primary component of the psychedelic brew ayahuasca [PMID:38868739, PMID:36275467]. It provides the potent hallucinogen N,N-dimethyltryptamine (DMT) to the decoction [PMID:29366418, PMID:31134518].

Background

Psychotria viridis, also known as chacrona, is a shrub in the coffee family Rubiaceae found in the Amazon region [PMID:34761362, PMID:36275467]. It is traditionally combined with the Banisteriopsis caapi vine to create ayahuasca, a psychoactive beverage [PMID:38868739, PMID:35449956].

Traditional uses

Indigenous shamans and native healers of the Orinoco and Amazon basins have traditionally used the brew for spirit communication, magical experiences, religious rituals, and as a healing tool to treat psychological disorders [PMID:29366418, PMID:31134518, PMID:39564645].

Active compounds

The primary active compound in Psychotria viridis is the psychedelic tryptamine N,N-Dimethyltryptamine (DMT) [PMID:34761362, PMID:36275467, PMID:26647632].

Mechanism of action

DMT acts as an agonist of the 5-HT2A receptor [PMID:31134518, PMID:26976063] and a partial agonist of 5-HT1A/2A receptors [PMID:39564645, PMID:25714595]. When combined with beta-carbolines from B. caapi, which inhibit monoamine oxidase-A (MAO-A), the systemic bioavailability of DMT is increased, allowing it to cross the blood-brain barrier and produce psychoactive effects [PMID:40839168, PMID:26165663].

Clinical evidence

Evidence BDepression

Open and randomized placebo-controlled clinical trials indicate significant and rapid antidepressant effects, starting as early as 1 day after intervention [PMID:34761362].

Evidence CAlcohol Use Disorder

Evidence suggests therapeutic potential for decreasing alcohol drinking and promoting abstinence [PMID:39523057, PMID:34822961].

Evidence DIschemic Stroke (Animal Model)

In Wistar rats, ayahuasca may attenuate ischemia-induced neuroinflammation and oxidative stress [PMID:40043852].

Safety & adverse effects

Potential risks include psychotic episodes related to DMT and potentially life-threatening serotonin syndrome [PMID:34060770]. Acute administration can cause moderate cardiovascular effects [PMID:25714595] and potent purging/emetic effects [PMID:34794194]. In a case report, ingestion by a patient with schizophrenia resulted in personal injury and property damage [PMID:29682363]. Chronic use in rats showed increased media thickness in the aorta [PMID:25714595].

Drug interactions

Consequences of use remain uncertain due to potential polydrug interactions [PMID:34060770].

Evidence summary

The evidence consists primarily of reviews and preclinical animal studies (Level C and D), with some encouraging results from clinical trials regarding depression (Level B).
