Boldo leaf (Peumus boldus) is a Chilean tree whose leaves contain the alkaloid boldine, the only natural compound with clinically recognized choleretic activity. It is primarily used as a liver tonic and for gallbladder disorders, with additional applications as a digestive aid and antioxidant. The herb's key active constituents include boldine, coumarin, volatile oils, and flavonoids.
Boldine stimulates bile secretion by acting on hepatocytes and increasing bile acid-dependent flow, likely via modulation of the farnesoid X receptor (FXR) and the bile salt export pump (BSEP). It also exhibits hepatoprotective effects through antioxidant activity, scavenging free radicals and reducing lipid peroxidation, as well as anti-inflammatory actions by inhibiting NF-κB and COX-2 pathways. Additionally, boldine may relax smooth muscle in the biliary tract, facilitating gallbladder emptying.
