Nigella Sativa Seed (Nigella sativa (seed only)): Benefits, Dosage, Safety & Evidence

Frequently asked questions

How does black seed work in the body?

Nigella sativa seeds work primarily through thymoquinone, their main active compound. Thymoquinone exerts antioxidant, anti-inflammatory, and immunomodulatory effects by modulating inflammatory pathways and supporting immune cell function. The seeds also contain fixed oils, proteins, and saponins that contribute to their therapeutic properties. Evidence level B supports these mechanisms, with research demonstrating effects on multiple biological pathways including NF-κB signaling and cytokine production.

Does black seed interact with blood thinners?

Yes, Nigella sativa seeds may interact with blood-thinning medications. The seeds contain compounds that can affect blood clotting and may enhance the effects of anticoagulants like warfarin, increasing bleeding risk. If you take blood thinners, consult your healthcare provider before using black seed. Additionally, black seed should be discontinued at least 2 weeks before scheduled surgery due to potential effects on blood clotting and anesthesia response.

How much black seed should I take daily?

The typical adult dosage for Nigella sativa seeds is 1-3 teaspoons daily. The seeds can be consumed whole, ground, or mixed with honey—a traditional method from Prophetic medicine. Start with smaller amounts (1 teaspoon) to assess tolerance, as the seeds can cause gastrointestinal irritation in some individuals. Consistency is important for therapeutic effects, but do not exceed recommended amounts without professional guidance.

Is black seed safe during pregnancy?

Nigella sativa seeds are considered unsafe during pregnancy, particularly in therapeutic amounts. The seeds contain compounds that may stimulate uterine contractions and affect pregnancy outcomes. While small culinary amounts used as a spice in Middle Eastern cooking may be acceptable, pregnant women should avoid medicinal doses entirely. Women who are breastfeeding should also consult a healthcare provider before using black seed supplements.

What are the health benefits of black seed?

Nigella sativa seeds have been used for centuries in Islamic Prophetic medicine and Egyptian traditional medicine for immune support and digestive health. Modern research (Evidence level B) suggests benefits for immune modulation, inflammatory conditions, and gastrointestinal health. The seeds are rich in thymoquinone, which provides antioxidant and anti-inflammatory properties. Traditionally, black seed has been called a remedy for numerous ailments in Prophetic tradition, highlighting its broad therapeutic reputation.

What are the side effects of taking black seed?

Common side effects of Nigella sativa seeds include gastrointestinal irritation, such as stomach upset, bloating, or nausea, particularly when taken on an empty stomach or in large amounts. Some individuals may experience allergic reactions. The seeds may also affect blood clotting, so people with bleeding disorders or scheduled surgery should exercise caution. Starting with small doses and taking seeds with food can help minimize digestive discomfort.

Summary

Nigella sativa, also known as black cumin or black seed, is a flowering plant native to Eastern Europe and the Middle East used in traditional medicine for various conditions [PMID:37184195]. It is recognized for its antioxidant, antimicrobial, and therapeutic efficacies [PMID:37184195].

Background

Nigella sativa is a flowering plant native to the Middle East and Eastern Europe that produces black seeds used for food flavoring and traditional medicine [PMID:37184195].

Traditional uses

It has been used in traditional medicine as a therapy for a wide variety of conditions [PMID:37184195] and has been traditionally valued for its medicinal properties [PMID:40905014]. In India, Türkiye, and the Middle East, it and the related species Nigella arvensis have been used orally as galactogogues [PMID:37184195].

Active compounds

The seeds contain an essential oil featuring terpenes, thymoquinone, and beta-elemene [PMID:37184195]. Phytochemical analysis shows high phenolic (35.48 mg GAE/g DW) and flavonoid (39.51 mg QE/g DW) contents, and fatty acids predominantly including methyl eicosatrienoate, methyl 11,14,17-eicosatrienoate, and methyl linoleate [PMID:40905014]. Thymoquinone is identified as a major bioactive component and the primary deodorizing substance [PMID:29437018, PMID:28701649].

Mechanism of action

Thymoquinone (TQ) modulates inflammation by preventing the biosynthesis of mediators such as 5-LO, COX, PGD2, and LTs, and reduces LPS-induced pro-inflammatory responses [PMID:29437018]. TQ may also inhibit cell proliferation and induce apoptosis by targeting epigenetic players including UHRF1, DNMT1, and HDAC1 [PMID:35566130]. Seed polysaccharides (NSSP) may regulate the PI3K and PTEN proteins and the PI3K-Akt signaling pathway [PMID:34166694].

Clinical evidence

Evidence BType 1 Diabetes Mellitus (Pediatric)

Administration of Nigella sativa seed oil (450 mg twice daily) for 3 months significantly lowered cholesterol, LDL-C, malondialdehyde, nitric oxide, TNF-α, and TGF-β [PMID:40048139].

Evidence DVitiligo

Topical application of a cream containing N. sativa seed oil for 6 months resulted in statistically significant repigmentation of the hands, face, and genital region [PMID:31025474].

Evidence BMenopausal Symptoms

A randomized placebo-controlled trial investigated the effects of N. sativa seed extract (910 mg/day and 1,365 mg/day) on estradiol, FSH, and vaginal maturity index in postmenopausal women [PMID:40352190].

Evidence DObesity-related conditions

Black cumin seed extract demonstrated lipid-lowering properties in human subjects and anti-adipogenic effects in 3T3-L1 cellular models [PMID:40905014].

Evidence CSinusitis

N. sativa seed fixed oil shows therapeutic potential via anti-inflammatory, antioxidant, antihistaminic, immune-modulator, antimicrobial, and analgesic effects, though clinical studies are required to evaluate efficacy in patients [PMID:29629288].

Safety & adverse effects

Black cumin is well tolerated, generally recognized as safe, and has not been associated with serum enzyme elevations or clinically apparent liver injury [PMID:37184195].

Pregnancy & lactation

Used orally as galactogogues in some regions, but no scientifically valid clinical trials support this use in humans; one study found a slight increase in serum prolactin but no evidence of growth promotion in infants [PMID:37184195].

Evidence summary

Evidence ranges from high-level randomized controlled trials for pediatric diabetes and menopause to preclinical cellular and animal models for cancer, BPH, and obesity. Review data strongly support its anti-inflammatory and antioxidant properties.
