Zizyphus Jujube Seed (Ziziphus jujuba var. spinosa): Benefits, Dosage, Safety & Evidence

Frequently asked questions

How does Zizyphus jujube seed work for sleep and anxiety?

Zizyphus jujube seed contains active compounds called jujubosides, along with betulin and oleic acid, which produce its calming effects. These compounds appear to modulate GABAergic neurotransmission and may influence serotonin pathways in the brain. In traditional Chinese medicine terms, the herb nourishes heart blood and calms the shen (spirit), addressing the root cause of insomnia and anxiety. The evidence level for these mechanisms is rated C, meaning proposed mechanisms are based on limited human data and traditional use rather than extensive clinical trials.

Does Zizyphus jujube seed interact with any medications?

Zizyphus jujube seed may potentiate the effects of central nervous system depressants, including benzodiazepines, barbiturates, and other sedative medications. Due to its sedative properties, combining it with alcohol or sleep medications may cause excessive drowsiness. There is limited evidence regarding specific drug interactions (Evidence Level C). Patients taking antidepressants, anxiolytics, or other CNS-active medications should consult a qualified healthcare practitioner before combining treatments. Always inform your healthcare provider about all supplements you are taking.

What is the recommended dosage of Zizyphus jujube seed for insomnia?

The typical adult dosage of Zizyphus jujube seed is 9-18 grams of seeds daily, traditionally prepared as a decoction or taken in powdered form. In clinical practice, it is often combined with other herbs in formulas tailored to the individual's pattern of disharmony. Lower doses may be appropriate when combined with other sedative herbs. This dosing range is based on traditional use and clinical experience (Evidence Level C). Always follow the specific recommendations of a qualified herbalist or healthcare provider for your individual needs.

Is Zizyphus jujube seed safe during pregnancy and breastfeeding?

Zizyphus jujube seed is generally considered safe during pregnancy when used at appropriate therapeutic doses under professional guidance. Traditional use has not identified specific concerns for pregnant or breastfeeding women. However, as with any herbal supplement during pregnancy, it should only be used when clearly indicated and under the supervision of a qualified healthcare practitioner. The safety rating is based on traditional use rather than extensive clinical trials (Evidence Level C). Consult your midwife or doctor before use.

What type of insomnia is Zizyphus jujube seed used for?

Zizyphus jujube seed is specifically indicated for insomnia associated with heart blood deficiency in traditional Chinese medicine. This pattern typically presents with difficulty falling asleep, vivid dreams, anxiety, palpitations, and possibly night sweats. It is not appropriate for insomnia caused by excess heat, which presents with restlessness, irritability, and feeling hot. The herb also addresses anxiety and spontaneous sweating. Evidence for these uses is rated Level C, based primarily on traditional use and limited clinical observation rather than rigorous controlled trials.

What are the side effects of Zizyphus jujube seed?

Zizyphus jujube seed is generally well-tolerated at recommended doses. The most commonly reported side effect is excessive drowsiness, particularly when taken in high doses or combined with other sedatives. Some individuals may experience digestive upset. The herb is contraindicated for insomnia caused by excess heat patterns, where it could worsen symptoms. Allergic reactions are possible but rare. Evidence for adverse effects is based on traditional use and clinical observation (Evidence Level C). Discontinue use if you experience unusual symptoms and consult a healthcare provider.

Summary

Zizyphus jujuba var. spinosa (Semen Ziziphi Spinosae), also known as Suan-Zao-Ren, is a traditional Chinese medicine primarily used for its sedative and hypnotic effects [PMID:36720435, PMID:41592108].

Background

Semen Ziziphi Spinosae (SZS) is the seed of Ziziphus jujuba var. spinosa (Bunge) Hu ex H.F. Chow, a plant widely distributed in China, Laos, Myanmar, and Iran [PMID:36720435]. It is recognized as a medicinal and edible resource [PMID:41302525].

Traditional uses

It is a classic traditional Chinese medicine used for treating insomnia [PMID:36720435, PMID:28084039, PMID:28899507], forgetfulness, and other nerve system diseases [PMID:32695210, PMID:37898440].

Active compounds

Identified bioactive compounds include jujuboside A and B [PMID:41584677], spinosin, mangiferin, neomangiferin, ferulic acid, liquiritin, isoliquiritin, and liquiritin apioside [PMID:28345817]. Overall, over 150 compounds have been identified, including flavonoids (164), triterpenoids (74), alkaloids, fatty acids, and polysaccharides [PMID:32695210, PMID:41592108].

Mechanism of action

The seeds modulate GABAergic activity and the serotonergic system [PMID:28899507], specifically by upregulating GABA receptor expression [PMID:41592108]. In animal models of Alzheimer's, Suan-Zao-Ren Decoction (SZRD) may ameliorate synaptic plasticity by inhibiting Aβ deposition and the JAK2/STAT3 signaling pathway [PMID:33478552].

Clinical evidence

Evidence BInsomnia (Methadone Maintenance)

Suan Zao Ren Tang significantly improved mean total PSQI scores and average sleep efficiency compared to placebo [PMID:26346534].

Evidence BInsomnia with Anxiety

A combination of Suan Zao Ren Tang and Zhi Zi Chi Tang significantly reduced ISI and PSQI scores and improved sleep architecture compared to lorazepam [PMID:25793006].

Evidence BInsomnia (Chronic Hepatitis B)

Modified Suan Zao Ren Decoction improved symptoms of insomnia and sleep stages (increased stage III, IV, and REM) compared to Surazepam and Diammonium Glycyrrhizinate [PMID:18323223].

Evidence DSleep difficulty (Climacteric women)

Suan Zao Ren Tang reduced PSQI total scores from 13.0 to 9.0, with superior benefit for daytime dysfunction in women with severe menopausal symptoms [PMID:21660310].

Evidence AGeneral Insomnia

A systematic review of RCTs found Chinese herbal medicine (CHM) to be similar to Western medication and placebo, but noted insufficient evidence due to poor methodological quality [PMID:22440393].

Safety & adverse effects

Reported adverse events associated with Suan Zao Ren Tang include dizziness, headache, stomach ache, and diarrhea [PMID:24159351]. A case of anaphylaxis was reported in a patient with pollinosis [PMID:39552699].

Evidence summary

Evidence for insomnia is mixed; while several clinical trials (B) show positive outcomes for specific populations, a systematic review (A) suggests overall evidence is insufficient due to low methodological quality of available RCTs.
