Bixa Orellana (Bixa orellana): Benefits, Dosage, Safety & Evidence

Questions fréquentes

How does Bixa orellana work as an anti-inflammatory?

Bixa orellana contains bioactive compounds including bixin, norbixin, and carotenoids that exert anti-inflammatory and antioxidant effects. Bixin, the primary carotenoid pigment, scavenges free radicals and inhibits pro-inflammatory mediators in the body. The flavonoids and tocotrienols contribute additional antioxidant activity. These compounds work synergistically to reduce oxidative stress and inflammation in tissues, which supports the traditional use for wound healing and inflammatory skin conditions. Evidence level: C (preliminary clinical evidence).

Does Bixa orellana interact with blood thinners?

Yes, Bixa orellana (annatto) may interact with anticoagulant and antiplatelet medications. The herb contains compounds that may affect blood clotting, potentially increasing bleeding risk when combined with warfarin, aspirin, clopidogrel, or other blood thinners. Additionally, annatto may lower blood sugar levels, which could enhance the effects of diabetes medications and cause hypoglycemia. Patients taking blood thinners or diabetes medications should consult a healthcare provider before using annatto supplements. Evidence level: C.

What is the recommended dosage for Bixa orellana?

Traditional preparations of Bixa orellana vary by application. For topical use, a seed paste can be applied directly to affected skin areas for wound healing and inflammatory skin conditions. For internal use as a tea, 5-10 grams of dried leaves daily is the traditional adult dosage. Standardized extracts are not well established. Always follow product-specific recommendations and consult a qualified herbalist or healthcare provider for personalized dosing, as optimal therapeutic doses have not been confirmed in clinical trials.

Is Bixa orellana safe during pregnancy?

Bixa orellana is considered unsafe during pregnancy and should be avoided. While annatto is commonly used as a food coloring in small amounts, medicinal doses may stimulate uterine contractions and potentially cause miscarriage or premature labor. High doses of annatto seeds have been associated with adverse pregnancy outcomes in traditional knowledge. Pregnant women, those planning pregnancy, and breastfeeding mothers should not use therapeutic doses of Bixa orellana. Food-level exposure through natural coloring is generally considered low risk.

What are the side effects of Bixa orellana?

Bixa orellana is generally well tolerated but can cause several side effects. The most common is orange-red skin staining from the bixin pigment when applied topically. Internal use may cause hypoglycemia (low blood sugar) in some individuals. Allergic reactions, though rare, can occur particularly in those sensitive to annatto. Symptoms may include skin rash, itching, or respiratory symptoms. People with known allergies to annatto food coloring should avoid medicinal preparations. Discontinue use if adverse reactions occur.

What is Bixa orellana used for medicinally?

Bixa orellana, commonly known as annatto or achiote, has been used traditionally for skin conditions, wound healing, fever reduction, and as an antimicrobial agent. The seeds contain bixin and other carotenoids with antioxidant properties that support skin health and tissue repair. Indigenous populations have used leaf preparations for fevers and infections. Modern research suggests potential benefits for inflammatory skin conditions, though clinical evidence remains limited (Evidence level C). The red pigment is widely used as a natural food coloring.

Résumé

Bixa orellana, commonly known as annatto or achiote, is a neotropical tree valued for its seeds and leaves, which exhibit antioxidant, antimicrobial, and anti-inflammatory properties [PMID:24357022, PMID:27708658].

Contexte

Bixa orellana (family Bixaceae) is a fast-growing perennial tree native to the neotropics [PMID:27708658]. It is widely used as a natural pigment (annatto) in the food, pharmaceutical, and cosmetic industries [PMID:27708658, PMID:29748818].

Usages traditionnels

Indigenous people have used leaf preparations for various medicinal applications [PMID:24357022]. It has been used in folkloric medicine to treat gonorrhea and other infections [PMID:24357022], and in Ayurvedic systems to treat dyslipidemia, diarrhoea, and hepatitis [PMID:37454777]. Other reported traditional applications include wound healing, analgesic, and hemostatic activities [PMID:34968663].

Composés actifs

The seeds contain high levels of carotenoids, primarily bixin (water-insoluble) and norbixin (water-soluble) [PMID:27708658, PMID:29748818]. Other active components include tocotrienols (specifically delta-tocotrienol), tocopherols [PMID:28559632, PMID:33124008], and a newly identified steroid called bixasteroid [PMID:34507510].

Mécanisme d'action

Bixin may ameliorate inflammation by inhibiting the TLR4/NF-κB, PI3K/Akt, and thioredoxin-interacting protein pathways [PMID:37781110], and by regulating Nrf2/MyD88/TLR4 and TGF-1/PPAR-/Smad3 pathways [PMID:37454777]. Delta-tocotrienol has been shown to protect against nicotine-induced DNA damage by regulating the PI3K/Akt-cyclin D1 pathway [PMID:30634632].

Preuves cliniques

Preuve BCardiovascular Risk

Consumption of eggs enriched with Annatto did not increase cardiovascular risk in healthy adults [PMID:36678239].

Preuve BGeneral Safety

A 6-month study in humans receiving 750 mg of leaf powder per day showed no significant or serious adverse effects [PMID:24357022].

Preuve DCutaneous Lesions (Animal Model)

A 10% aqueous extract gel demonstrated skin healing potential in exposed cutaneous lesions in rats [PMID:37213245].

Preuve DAntifungal (In Vitro)

Ethanol extracts of Bixa orellana exhibited antifungal activity against Candida albicans [PMID:31883250].

Sécurité et effets indésirables

Human data indicates that 750 mg of leaf powder per day for 6 months is safe with no serious adverse effects [PMID:24357022]. In rats, a 90-day repeated-dose oral toxicity study of geranylgeraniol (from annatto oil) showed no evidence of mutagenicity or genotoxic activity, though treatment-related adverse effects were observed at certain doses [PMID:34144118].

Synthèse des preuves

The evidence consists primarily of in vitro studies, animal models, and a few reviews (Level C/D), with limited human clinical data (Level B) focusing on safety and cardiovascular neutrality.
