Devil's Club (Oplopanax horridus): Benefits, Dosage, Safety & Evidence

Questions fréquentes

How does Devil's Club work in the body?

Devil's Club contains several bioactive compounds, including saponins, polysaccharides, and alkaloids, which are believed to contribute to its medicinal effects. The saponins may help modulate immune function and exert anti-inflammatory activity, while polysaccharides could support blood sugar regulation. However, the exact mechanism of action remains poorly understood, as most evidence comes from traditional use rather than clinical trials (Evidence Level C). More research is needed to fully understand how Devil's Club produces its adaptogenic and metabolic effects.

Does Devil's Club interact with diabetes medications?

Yes, Devil's Club may interact with diabetes medications. Traditional use and preliminary evidence suggest it may help lower blood sugar levels, which could enhance the effects of antidiabetic drugs like metformin or insulin. This combination may cause blood sugar to drop too low (hypoglycemia), resulting in dizziness, weakness, or confusion. If you take diabetes medications, consult your healthcare provider before using Devil's Club, and monitor blood glucose levels closely (Evidence Level C).

What is the recommended dosage for Devil's Club?

Traditional adult dosages for Devil's Club include 2-4 mL of tincture (1:5 strength) taken three times daily, or 2-4 grams of dried bark prepared as a decoction. These recommendations are based on traditional use by Pacific Northwest Indigenous peoples rather than standardized clinical trials (Evidence Level C). Start with lower doses to assess tolerance, and consult a qualified healthcare practitioner for personalized guidance, especially if you have underlying health conditions.

Is Devil's Club safe during pregnancy and breastfeeding?

No, Devil's Club is considered unsafe during pregnancy and breastfeeding. Traditional knowledge and its potential effects on blood sugar and uterine function suggest it should be avoided during these periods. There is insufficient safety data for use during lactation. Women who are pregnant, trying to conceive, or breastfeeding should not use Devil's Club. Always consult your healthcare provider before using any herbal supplement during pregnancy (Evidence Level C, Traditional).

What is Devil's Club used for traditionally?

Devil's Club has been used for centuries by Pacific Northwest Indigenous peoples as one of their most sacred medicinal plants. Traditional applications include managing diabetes and blood sugar, treating rheumatism and arthritis, addressing respiratory infections, and serving as an adaptogen to help the body resist stress. The root and inner bark are typically prepared as teas, decoctions, or tinctures. These uses are based on extensive traditional knowledge rather than modern clinical trials (Evidence Level C, Traditional).

What are the side effects of Devil's Club?

Devil's Club can cause several side effects. The plant's formidable spines may cause skin irritation and injury during handling, so wear protective gloves when preparing it. Internal use may cause gastrointestinal upset in some individuals. Because Devil's Club may lower blood sugar, hypoglycemia is a potential concern, especially for those with diabetes. Discontinue use if you experience dizziness, weakness, or other signs of low blood sugar (Evidence Level C).

Résumé

Devil's Club (Oplopanax horridus) is a medicinal plant native to North America, particularly the Pacific Northwest, used traditionally for various ailments and currently studied for its anticancer and anti-inflammatory properties [PMID:30302120, PMID:20723598].

Contexte

Oplopanax horridus is a botanical native to North America and is the only species of the genus Oplopanax distributed in this region [PMID:22101399]. It is considered a significant spiritual and medicinal plant for indigenous peoples of Alaska and the Pacific Northwest [PMID:20723598, PMID:25340187].

Usages traditionnels

Traditionally used by indigenous peoples for internal and external infections, arthritis, respiratory ailments, digestive tract ailments, broken bones, fever, headaches, cancer, colds, cough, and diabetes [PMID:20723598, PMID:15154981, PMID:16814500].

Composés actifs

Identified compounds include polyynes (such as falcarindiol and oplopantriol A and B), polyenes (including (S,E)-nerolidol), sesquiterpenes, monoterpenoids, phenylpropanoids, phenolic acids, lignans, glycosides, and lupane-type triterpenoid saponins [PMID:22101399, PMID:32496571, PMID:20734931, PMID:25282894, PMID:20638214].

Mécanisme d'action

Polyacetylenes have been shown to activate PPARγ, a transcription factor involved in metabolic regulation [PMID:32129622]. In cancer cells, compounds modulate genes related to pro-apoptosis, anti-apoptosis, cell cycle, stress, and death receptors [PMID:30661403]. Leaf extracts reduce LPS-induced nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) production by decreasing iNOS expression [PMID:30263531].

Preuves cliniques

Preuve DCancer (Various types)

In vitro studies demonstrate antiproliferation activity against breast, colon, pancreatic, ovarian, and leukemia cell lines [PMID:20723598, PMID:24215675, PMID:19835944, PMID:26221768, PMID:25340187, PMID:20332432]. Oplopantriol A showed dose-related suppression of tumor growth in xenograft colon cancer mouse models [PMID:25045937].

Preuve DDiabetes Mellitus

A pilot study monitoring blood glucose levels in insulin-dependent and non-insulin-dependent diabetic patients showed no hypoglycemic effect of devil's club tea [PMID:21249104].

Preuve DInflammation

Leaf extracts demonstrated anti-inflammatory effects by reducing LPS-induced NO, TNF-α, and IL-6 in murine macrophages [PMID:30263531].

Preuve DMycobacterial Infections

Isolated polyynes exhibited significant antibacterial and antimycobacterial activity, including the ability to kill Mycobacterium tuberculosis and isoniazid-resistant Mycobacterium avium [PMID:9392889].

Interactions médicamenteuses

In vitro studies suggest that Devil's club extract may enhance the antiproliferation activity of chemotherapeutic agents such as cisplatin (CDDP) and gemcitabine (GEM) [PMID:24215675, PMID:26221768].

Synthèse des preuves

The current evidence is primarily based on in vitro cell studies, animal models (xenografts), and limited pilot observations. There is a lack of high-level clinical trial data (RCTs) or systematic reviews regarding human efficacy and safety.
