Centella asiatica (Gotu Kola) is a traditional wound-healing herb whose topical preparations are widely used to accelerate wound closure, stimulate collagen synthesis, and reduce scar formation. Its key active triterpenoids—asiaticoside, madecassoside, asiatic acid, and madecassic acid—modulate fibroblast activity and inflammatory mediators.
Topical Centella asiatica promotes wound healing primarily through the upregulation of collagen type I and III synthesis via activation of TGF-β/Smad signaling pathways. Asiaticoside enhances fibroblast proliferation and migration, while madecassoside inhibits pro-inflammatory cytokines such as TNF-α and IL-6. The triterpenoids also increase angiogenesis by upregulating VEGF and improve wound tensile strength. Additionally, asiatic acid exhibits antioxidant activity by scavenging ROS and modulating the Nrf2 pathway.
