Magnolia grandiflora, a North American ornamental tree, has bark and leaves used traditionally for fever and rheumatism. Modern research indicates antipyretic, anti-inflammatory, and anxiolytic properties, attributed primarily to the neolignans magnolol and honokiol. Evidence is limited (Level C) and primarily from in vitro and animal studies.
Magnolol and honokiol exert anti-inflammatory effects via inhibition of COX-2, 5-LOX, and NF-κB pathways. Anxiolytic effects are mediated through modulation of GABA-A receptors, similar to benzodiazepines but with less sedation. Additionally, these compounds show antioxidant and antimicrobial activities. The antipyretic action may involve central prostaglandin inhibition.
