Usnea Lichen (Usnea barbata): Benefits, Dosage, Safety & Evidence

Questions fréquentes

How does usnea lichen work as an antimicrobial?

Usnea lichen works primarily through usnic acid, which disrupts bacterial cell membranes and inhibits protein synthesis. This mechanism makes it effective against Gram-positive bacteria like Streptococcus and Staphylococcus species, as well as some Gram-negative bacteria and fungi. The lichen also contains diffractic acid and polyphenols that contribute to antimicrobial activity. Traditional use across European, Chinese, and Native American medicine supports these antimicrobial properties for wound healing and respiratory infections. Evidence level: C.

Does usnea lichen interact with medications?

Usnea may interact with hepatotoxic medications, including acetaminophen, statins, and antifungal drugs, due to its potential for liver toxicity. Combining usnea with alcohol or other liver-stressing substances increases hepatotoxicity risk significantly. Usnea may also enhance effects of anticoagulant medications. Those taking prescription medications should consult a healthcare provider before using usnea supplements. Evidence level: C. Always disclose supplement use to your healthcare provider.

What is the recommended dosage for usnea lichen?

Standard adult dosages include 300mg of usnic acid standardized extract twice daily, or 3-5ml of tincture three times daily. For topical wound healing, usnea can be applied as a poultice, salve, or infused oil. Internal use should be limited to short durations due to hepatotoxicity concerns. Those with liver conditions should avoid internal use entirely. Evidence level: C. Consult a qualified medical herbalist for personalized dosing recommendations.

Is usnea lichen safe during pregnancy and breastfeeding?

Usnea is considered unsafe during pregnancy and breastfeeding. The usnic acid content poses hepatotoxicity risks, and insufficient safety data exists for fetal or infant exposure. Pregnant and nursing individuals should avoid all usnea supplements. While traditional use included topical wound care, internal consumption during pregnancy has not been established as safe. Evidence level: D (expert opinion based on constituent toxicity). Seek guidance from a qualified healthcare provider for pregnancy-safe alternatives.

What are the side effects of usnea lichen?

The primary side effect of usnea is hepatotoxicity, particularly with high internal doses of usnic acid. Reported cases include elevated liver enzymes, jaundice, fatigue, and in severe instances, liver failure requiring transplantation. Those with pre-existing liver disease should avoid usnea completely. Topical application is generally well-tolerated with minimal side effects. Internal use should be limited to short durations under professional supervision. Evidence level: C. Discontinue use if experiencing unexplained fatigue or yellowing of skin.

What is usnea lichen used for in herbal medicine?

Usnea lichen is used traditionally for wound healing, antisepsis, respiratory infections, and urinary tract infections. European folk medicine specifically employed usnea for strep throat and bronchitis. The antimicrobial properties come primarily from usnic acid, effective against bacteria and fungi. Modern herbalists use usnea for immune support and as a natural antibiotic alternative. Evidence level: C (preliminary clinical evidence and traditional use). Topical applications remain the safest use due to hepatotoxicity concerns with internal doses.

Résumé

Usnea sp. is a fruticose thalli lichen recognized for its multidirectional biological effects, most notably its antimicrobial activity attributed to usnic acid [PMID:33276059].

Contexte

Usnea barbata (also known as 'old man's beard') is a fruticose lichen that produces various bioactive metabolites, including polyketides and polyphenols [PMID:32878015, PMID:26745885].

Usages traditionnels

Usnea sp. has been used in traditional medicine worldwide since ancient times to treat various diseases [PMID:33276059]. In Romania, U. barbata is known by popular names such as 'Bear's beard', 'Trees' dandruff', and 'Woman's long hair' [PMID:7982010].

Composés actifs

The most prominent secondary metabolite is usnic acid [PMID:19034791, PMID:32316674]. Other bioactive components include phenolic compounds, flavonoids, and unique polyketides [PMID:29596056, PMID:32878015].

Mécanisme d'action

Anticancer effects are associated with the induction of apoptosis, autophagy, and the production of reactive oxygen species (ROS), which leads to DNA damage [PMID:26745885, PMID:36499160, PMID:32756347]. Anti-inflammatory effects in keratinocytes involve the inhibition of prostaglandin E2 (PGE2) production without affecting COX-2 protein expression [PMID:17766140].

Preuves cliniques

Preuve DBacterial and Fungal Infections

Extracts exhibit antimicrobial activity against Gram-positive (e.g., S. aureus, E. faecalis) and Gram-negative (e.g., P. aeruginosa, E. coli) bacteria, as well as Candida species [PMID:32878015, PMID:36009320, PMID:35009036, PMID:42076002].

Preuve DOral Squamous Cell Carcinoma (OSCC)

Methanol and oil extracts induce cell killing, apoptosis, and DNA damage in OSCC cell lines (e.g., CAL-27, Ca9-22) via oxidative stress [PMID:32756347, PMID:36499160, PMID:34356377].

Preuve DInflammation and Pain

Hydroalcoholic extracts demonstrated anti-inflammatory activity comparable or superior to phenylbutazone and hydrocortisone hemisuccinate, and analgesic activity close to noraminophenazone [PMID:7982010].

Preuve DUVB-induced Skin Inflammation

U. barbata extract inhibits PGE2 production in HaCaT keratinocytes, suggesting protective effects against UVB-mediated inflammatory skin conditions [PMID:17766140].

Sécurité et effets indésirables

Usnic acid has been linked to hepatotoxicity; the US FDA received 21 reports of liver toxicity related to dietary supplements containing usnic acid, leading to a warning in 2001 [PMID:19034791, PMID:38904414]. Acute toxicity studies in animals showed LD50 values for U. barbata at 22.53g/kg (i.p.) and 7.43g/kg (i.v.), with no mortality recorded per os up to 32g/kg [PMID:7982010].

Synthèse des preuves

The current evidence is primarily based on in vitro studies (cell lines), animal models (fish, brine shrimp), and review articles. There is a lack of human clinical trials (RCTs), resulting in a low overall strength of clinical evidence (Level D).
