Brucea javanica (Ya Dan Zi) is a Southeast Asian medicinal plant from the Simaroubaceae family, traditionally used in TCM for dysentery, malaria, and tumors. Its key active compounds are quassinoids (bruceosides, brusatol) which exhibit anti-tumor, anti-malarial, and anti-protozoal activities. Modern research focuses on its cytotoxic and anti-inflammatory properties, though clinical evidence remains limited (grade C).
Brucea javanica quassinoids, particularly bruceosides and brusatol, exert anti-tumor effects by inhibiting protein synthesis via ribosomal inactivation and inducing apoptosis through NF-κB pathway suppression and Nrf2 inhibition. Anti-malarial activity involves disruption of Plasmodium falciparum mitochondrial function and inhibition of heme polymerization.
